Overview
MOTS-C (mitochondrial open reading frame of the 12S rRNA-c) is a mitochondrial-derived peptide composed of 16 amino acids. It is encoded within mitochondrial DNA and has been identified in several tissues as well as circulating plasma. Because of its ability to function both within cells and extracellularly, researchers have studied MOTS-C as part of a group of signaling molecules sometimes referred to as mitochondrial-derived peptides (MDPs).
Scientific studies have examined MOTS-C for its potential interaction with AMP-activated protein kinase (AMPK) signaling pathways, which play a key role in cellular energy sensing and metabolic regulation. Under conditions of metabolic stress, researchers have suggested that MOTS-C may translocate from the mitochondria to the cell nucleus, where it may influence gene expression involved in metabolic homeostasis.
Because of these properties, MOTS-C has been studied as a model compound in investigations exploring mitochondrial communication, metabolic signaling pathways, and cellular stress responses.
Research Background
Experimental research has explored MOTS-C in studies evaluating metabolic regulation, skeletal muscle physiology, and cellular energy balance.
In animal research models, investigators observed that exposure to MOTS-C appeared to influence AMPK activation and glucose metabolism within skeletal muscle tissue. Researchers have suggested that these interactions may affect glucose transporter expression and cellular nutrient utilization pathways involved in energy metabolism.
Additional studies have examined the peptide’s interaction with mitonuclear signaling, where mitochondrial-encoded peptides may influence nuclear gene expression. Researchers have reported that MOTS-C may regulate genes associated with antioxidant response elements and stress-responsive transcription pathways, suggesting a role in cellular adaptation to metabolic stress.
Research has also investigated the peptide in models examining fat metabolism, bone signaling pathways, and endothelial cell function. Some studies have suggested interactions with pathways such as TGF-β/SMAD signaling in bone cells and potential influences on vascular endothelial responses through AMPK-related signaling.
Because of its role in mitochondrial signaling and metabolic regulation, MOTS-C has also been examined in studies exploring cellular aging, energy metabolism, and exercise-related metabolic responses.
